چکیده

In this research paper, we provide an introduction to fluorescence microscopy techniques. We show how the development of these techniques has lead to significant advances in cellular physiology. As part of this discussion, we detail applications of optical imaging to a study of cell division and to a study of the nervous system. We will conclude by presenting some limitations and controversies of these techniques, and will also outline some exciting new developments for the future.

Comparative Studies of High Contrast Fluorescence Imaging Efficiency of Silica-coated CdSe Quantum Dots with Green and Red Emission

Herein we report the possibility of using green and red emitting silica-coated cadmium selenide (CdSe) quantum dots (QDs) for remarkable stem and cancer cellular imaging, efficient cellular uptake and fluorescence imaging of semi and ultra-thin sections of tumor for in vivo tumor targeted imaging applications. The comparative studies of high contrast cellular imaging behaviours of the silica-co...

Dual-modality fluorescence and full-field optical coherence microscopy for biomedical imaging applications

Full-field optical coherence microscopy (FFOCM) is a high-resolution interferometric technique that is particularly attractive for biomedical imaging. Here we show that combining it with structured illumination fluorescence microscopy on one platform can increase its versatility since it enables co-localized registration of optically sectioned reflectance and fluorescence images. To demonstrate...

A Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...

Principles of Two-Photon Excitation Microscopy and Its Applications to Neuroscience

The brain is complex and dynamic. The spatial scales of interest to the neurobiologist range from individual synapses (approximately 1 microm) to neural circuits (centimeters); the timescales range from the flickering of channels (less than a millisecond) to long-term memory (years). Remarkably, fluorescence microscopy has the potential to revolutionize research on all of these spatial and temp...

Two-photon Scanning Fluorescence Microscopy under Total Internal Reflection

Two-photon fluorescence microscopy is a powerful method for many applications. Because of the ability of strong attenuation of the background fluorescence, total internal reflection fluorescence microscopy (TIRFM) has been the major technique in the fluorescence imaging and single molecule detections. Recently, we have developed a new type of TIRFM called scanning TIRFM (henceforth abbreviated ...
